1. I don't think it is nicknameish because it is a name in its own right.
2. I think it works fine as a nickname for Margaret or Marguerite.
3. I used to work with a Daisy. She is probably in her early 20s now.
4. I don't like any other spellings.
5. Daisy Charlotte is pretty. Other suggestions:
